天然免疫赋予机体对病原及肿瘤细胞的抵抗力,是人类抵抗感染与肿瘤的第一道防线。天然免疫失调引起各种感染性疾病、自身免疫病和肿瘤。尤其重要的是,识别细胞内DNA的cGAS-STING通路,对肿瘤的免疫监视及免疫治疗都至关重要。蒋争凡实验室是世界上同期发现STING蛋白的三个实验室之一,对其生理学功能进行了深入系统的研究。发现ERIS的二聚体化对其激活天然免疫反应重要;细胞通过STING-TBK1活化STAT6及NF-kB在cGAS-STING通路中的重要作用;发现特异性降解3'3'-cGAMP的磷酸二酯酶;发现多种Caspase负调控天然免疫反应,避免过度的免疫反应及凋亡细胞保持“免疫沉默”的机制;发现锰元素在cGAS-STING通路中发挥“警报素”与“激动剂”的双重功能,以此为基础发现锰离子的免疫调节功能,为锰元素在体内的生理学功能,特别是作为免疫调节剂的功能研究打开了一个新的窗口,并揭示锰元素未来在抗感染、抗肿瘤及免疫佐剂等应用方面拥有广阔的前景。
